Recently, we published a paper on screening neutral sites for metabolic engineering of Ogataea polymorpha and realized fatty alcohol synthesis via this system (Synthetic and Systems Biotechnology, 2021, 6, 63-68).
O. polymorpha is capable to utilize methanol, making it a promising chassis cell. However, lack of genome integration sites limited its extensive metabolic engineering. Therefore, we systematically identified 17 neutral sites suitable for gene integration without compromising cell fitness, among which 3 neutral sites were recognized as high expression locus. In addition, fatty alcohol biosynthesis was realized by integration of corresponding genes in Ogataea polymorpha. (Text and image by Wei Yu)
