Recently, our group engineered Pichia pastoris for bioproduction of 3‐hydroxypropionic acid (3-HP) from sole methanol.
3-HP is a versatile platform chemical for the synthesis of various biodegradable polymeric materials. In this study, we optimized the expression of malonyl-CoA reductase gene MCR. Then, we metabolically transformed a free fatty acid (FFA) overproducing strain toward 3-HP biosynthesis. Finally, downregulating of methanol dissimilative pathway further improved 3-HP production to 48.2 g/L with a yield of 0.23 g/g under fed-batch fermentation in bioreactor. This is the highest 3-HP production from one-carbon (C1) feedstocks reported so far.
